Following the ascension to power of Taira no Kiyomori, the Minamoto clan is exiled. Minamoto no Yoritomo meets Masako, the sister of Hōjō Yoshitoki and later marries her. Following this marriage, the gears of Yoshitoki's destiny begin to turn.

Set in the 18th century, the show follows the Shogun Yoshimune, who likes to disguise himself as a low-ranking samurai and go into his capital of Edo to see the life of the common man, as well as to seek out and punish evildoers who would hurt his citizens. He is aided by Magistrate Oo'oka and a vivacious fireman, Tatsugoro, as well as a rotating cast of other recurring characters. Along with Zenigata Heiji and Mito Kōmon, it ranks among the longest-running series in the jidaigeki genre. Like so many other jidaigeki, it falls in the category of kanzen-chōaku, loosely, "rewarding good and punishing evil."

A new shogun is appointed to the throne when his brother dies. He find out that the former Shogun one of the women from his brothers harem became pregnant, but she escaped. The new Shogun would loose the throne if the child is a boy. Genshiro is doctor who, among other things, performs abortions, and is sheltering his love, another women who escaped from the harem. The new Shogun threatens to arrest the girl unless Genshiro hunts down the woman and aborts her child. Genshiro learns that the woman releases a "musky scent from her special area" when in ecstasy. Both Genshiro and his friend set out to find all the women of the former harem and have sex with them, by force if necessary.

Tomizo, a master storehouse-breaker who has been released from prison in Denmacho after serving five years plans to break into the Edo Castle gold storehouse to avenge the death of his father, who died a violent death at a gold mine on Sado Island. With the help of Hanji, a playboy who took care of him in prison, as his partner, Tomizo decides on the night of fireworks to be held in Edo Castle as the day to carry out his plan. Samon Kamiyama, a yoriki of the Minami-machi magistrate infiltrates the Denzū-machi prison disguised as Mushuku Sahēji. Samon is mistaken for a snitch and about to be beaten up when he was saved by the quick thinking of a ferryman Sanji. The infiltration is the plan of Samon and chief elder Abe Isenokami, who has asked Samon to obtain smuggling evidence against shipping whole retailers Kawachiya Senemon and his son Sennosuke.

In the year 1716 a mysterious epidemic stickens men in the country of Japan, dropping the population of men to 1/4th of its prior state. With the drastic reduction of men, the gender roles have become reversed in Japan. Woman become the dominant members of society and males are sought out for the ability to produce children. A young man named Unoshin Mizuno hopes to marry childhood sweetheart Onobu, but due to class differences realizes this is almost impossible. To raise his social status and also save his poor family, Unoshin Mizuno enters the Ohoku (inner chambers of the Shugun's castle) and attempts to vie among 3,000 other men for the affection of the female shogun. What Unoshin Mizuno quickly learns about the Ohoku is that the men there are all beautiful, but highly ambitious and conniving. In this environment, the 7th shogun Tokugawa passes away and the new shogun Yoshimune Tokugawa takes the thrown and enters the Ohoku.

From the opening moments when prisoners carry out their own form of justice, killing a crooked ex-constable, it is clear that this is the work of a visionary filmmaker.
